Untargeted Lipidomics Analysis

Lipidomics is a discovery-based research approach focusing on detection and relative quantitation of known and unknown lipids in a given biological system. Lipidomics is a newly emerged discipline that studies cellular lipids on a large scale based on analytical chemistry principles and technological tools, particularly mass spectrometry. Lipidomic analysis can detect lipid classes in blood with high-resolution devices.

 

Untargeted lipidomics makes it possible to analyze hundreds to thousands of individual lipid species simultaneously, which is valuable to evaluate an individual’s health status. Differences in lipid profiles have been reported in investigations of cancer, diabetes, Alzheimer’s disease and cardiovascular disease. Such amounts of detailed lipid profiles are available to assess medical risks, monitor and optimize patient treatments and are the basis for the concept of precision medicine. 

Service summary

The LC-MS technology involves use of an UHPLC, wherein the individual components in a mixture are first separated followed by ionization and separation of the ions on the basis of their mass/charge (m/z) ratio. LC-MS/MS based Orbitrap technology is used in the Lipidomics test.   The development of HRAM mass spectrometry has risen to the challenge of lipid analysis and offers an unprecedented level of sensitivity, selectivity and precision.

 

  • Literature based sample preparation with Avanti Splash Lipidomix Mass Spec standard solutions from transferred samples (homogenizing, lipid quenching, extracting, reconstituting)
  • Analyzing with Thermo Scientific Orbitrap Exploris 240 MS system
  • Lipid identification using Lipid Search and Compound Discoverer software
  • Data processing via Compound Discoverer 3.3 software
  • Turnover time approx. 2 to 3 weeks**

After the plasma is separated, it is taken into the appropriate tube and centrifuged at 13000 × g, 15 min, 4 °C. After centrifugation, supernatant is portioned into at least 2 tubes (or more) with 100 µl. Stored at −80 °C until analysis.

What we do with your samples

Once your samples arrive at Phenome Omics, we start by performing a reception control step in which we make sure the samples meet our requirements.

 

If the samples fail this quality control step, we will contact you to discuss possible options. If the samples pass reception control, we will inform you and the samples will be queued for sample preparations.

 

In our laboratory, sample preparation, lipid identification and data analysis for global lipidomics are performed with state-of-the-art systems. We aim to acquire as much lipid information as possibble from just a few microliters of sample material. 

 

We use Thermo Scientific Orbitrap Exploris 240 MS for Untargeted Lipidomics Analysis. 

 

This system offers the following qualifications:

 

·       Exceptional mass accuracy 

·       Ultra-high resolution up to 240k 

·       Regular & Mild trapping 

·       Extended low mass to m/z 40 

·       Robust instrument performance 

·       AcquireX intelligent acquisition 

 

By means of this technology we are able to use AcquireX Deep Scan for low abundance compounds. 

 

After the lipidomics data is acquired from mass spectrometry, Lipid Search and Compound Discoverer 3.3 software are used for data processing. Through the instrumentality of Compound Discoverer software, feature detection with integrated databases & spectral library, data reduction, unknown annotations, statistics, pathway mappings are able to carry out.

Epidemiological studies have provided much insight into the relationship between plasma lipid profiles, health and disease.
